A Convenient U-Shape Microreactor for Continuous Flow Biocatalysis with Enzyme-Coated Magnetic Nanoparticles-Lipase-Catalyzed Enantiomer Selective Acylation of 4-(Morpholin-4-yl)butan-2-ol

This study implements a convenient microreactor for biocatalysis with enzymes immobilized on magnetic nanoparticles (MNPs). The enzyme onto MNPs by adsorption or covalent bonds was lipase B from Candida antarctica (CaLB). were obtained covering the magnetite core silica shell and later hexadecyltrimethoxysilane, while immobilization, silica-covered functionalized layer forming mixtures of hexadecyl- 3-(2-aminoethylamino)propyldimethoxymethylsilanes in 16:1 molar ratio, which further activated neopentyl glycol diglycidyl ether (NGDE). resulting CaLB-MNPs tested continuous flow system, created 3D printing to hold six adjustable permanent magnets beneath polytetrafluoroethylene tube (PTFE) anchor MNP biocatalyst inside reactor. anchored formed reaction chambers passing fluid through above biocatalysts, thus increasing mixing during enhanced activity CaLB MNPs. enantiomer selective acylation 4-(morpholin-4-yl)butan-2-ol (±)-1, being chiral alcohol constituent mucolytic drug Fedrilate, carried out U-shape reactor compared batch reactions lyophilized using same composition, amounts showed similar higher mode superior as powder form. magnet design represents general easy-to-access implementation MNP-based microreactors, useful many biotransformations reducing costly time-consuming downstream processes.
